Description
Rozlytrek, or Entrectinib, is a medicine used to treat specific types of cancer, including ROS1-positive non-small cell lung cancer and solid tumors with NTRK gene fusions. It works by blocking certain proteins that help tumors grow. Rozlytrek comes in the form of capsules, which patients take by mouth. By targeting these specific gene changes, it helps slow down or stop the growth of cancer cells in affected patients.
Directions
Rozlytrek is typically taken once daily, either as capsules or oral pellets. It’s important to swallow the capsules whole; do not crush, chew, or dissolve them. For oral pellets, follow the specific mixing instructions provided by your doctor. Adults usually start with a standard dosage of 600 mg per day. Children’s dosage depends on their size and weight.

- Rozlytrek may raise your uric acid levels, which can lead to gout. Watch for symptoms like sudden joint pain, especially in the big toe, and inform your doctor if they occur.
- Rozlytrek can affect your heart's rhythm by prolonging the QT interval. If you have a history of heart rhythm issues or are taking other medications that affect your heart, discuss this with your doctor before starting Rozlytrek.
- Rozlytrek might cause changes in your vision, such as blurred vision or sensitivity to light. If you experience any vision changes, stop taking Rozlytrek and seek medical advice immediately.
- Rozlytrek can harm an unborn baby. If you are pregnant or planning to become pregnant, use effective contraception during treatment and for five weeks after the last dose. Men with female partners who can become pregnant should use contraception during treatment and for three months after the last dose.
Side Effects
The most common side effects of Rozlytrek include tiredness, nausea, and weight gain. You may also experience constipation, muscle and joint pain, dizziness, and changes in taste or vision. Some people report confusion, memory problems, hallucinations, or other changes in mental status. Other side effects can include cough, shortness of breath, vomiting, and swelling.
Rozlytrek can cause serious side effects like congestive heart failure, leading to symptoms such as shortness of breath, swelling in the legs, and fatigue. It may also affect your central nervous system, causing dizziness, mood changes, or confusion. Bone fractures and liver problems have also been reported. If you notice signs of high uric acid levels, such as joint pain or pink urine, or if you experience irregular heartbeats or vision changes, contact your healthcare provider immediately.
