Description
Piqray (alpelisib) is an oral medication prescribed to treat adults with:
- hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative, PIK3CA-mutated, breast cancer that has advanced or spread to other parts of the body and has progressed during or after receiving endocrine-based treatment
Piqray is available in 50 mg, 150 mg, and 200 mg tablets.

Directions
Take Piqray exactly as prescribed by your doctor. Carefully read the medication guide that comes with your prescription.
Piqray dosing:
Your doctor will tell you how much Piqray you should take. The recommended dose is 300 mg by mouth one time a day with food.
Piqray storage:
Keep Piqray tablets at room temperature between 68°F and 77°F (20°C and 25°C).
Keep the medication out of reach of children.

Ingredients
The active ingredient in Piqray is alpelisib.
Inactive ingredients include hypromellose, magnesium stearate, mannitol, microcrystalline cellulose, and sodium starch glycolate. The film-coating contains hypromellose, iron oxide black, iron oxide red, macrogol/polyethylene glycol (PEG) 4000, talc, and titanium dioxide.
Contraindications
Patients with an allergy to Piqray or to any of its ingredients should not take it.
Cautions
- Before you begin taking Piqray, talk to your doctor about:
- All the prescription and over-the-counter medications you take
- Your allergies
- Your current health problems and past medical history
- Your pregnancy or breastfeeding status
- Piqray is associated with the following warnings and precautions:
- Severe hypersensitivity reactions
- Severe cutaneous adverse reactions
- High blood glucose
- Pneumonitis and interstitial lung disease
- Diarrhea or colitis
- Harm to an unborn baby. Effective contraception is recommended for females of childbearing age and males with female partners who are of childbearing age during treatment with Piqray and for one week after taking the final dose.
- Breastfeeding is not recommended.
Side Effects
High blood glucose; elevated creatinine; diarrhea; rash; decreased blood glucose, lymphocytes, and hemoglobin; increased gamma-glutamyl transferase (GGT), liver enzymes, and lipase; nausea; fatigue; decreased appetite; swelling/sores of the mouth; vomiting; weight loss; decreased calcium levels; hair loss; and prolonged blood clotting time are common Piqray side effects. However, other side effects may also occur. Immediately tell your doctor about any side effects that bother you or won’t go away.
